Columns for purification of Strep-tag proteins under gravity flow


Gravity flow column with 0.2 ml Strep-Tactin resin. Gravity flow column with 1 ml Strep-Tactin resin. Also available with Ni-NTA resin. Gravity flow columns with 5 ml and 10 ml Strep-Tactin resin; 5 ml column also available with Ni-NTA resin. Note: The gravity flow columns illustrated here are pre-packed with Strep-Tactin Sepharose

Our Column Evaluation Sets enable the comparison of the three different resins. Available for two gravity flow column sizes.

Handling and regeneration of Strep-Tactin gravity flow columns

Gravity flow columns cannot run dry allowing a step-by-step purification procedure without the need for further equipment. After purification, columns are regenerated with a yellow regeneration buffer containing 1 mM HABA (2-[4'-hydroxy-benzeneazo] benzoic acid. The regeneration process on the column can be followed visually by color change from yellow to red. Prior to the next run the dye is washed off with fast kinetics by equilibration buffer. Please note, that MacroPrep and POROS* do not turn as red as Sepharose (see Strep-Tactin Resins-Specifications) and may keep a slight pink color after regeneration. Evaluation Set
Strep-Tactin columns regenerated with HABA (see also Strep-tag purification cycle):
Strep-Tactin MacroPrep, Superflow and Sepharose (left to right). Since MacroPrep is not as transparent as Sepharose or Superflow, the color shift to red is less visible. Scale: reduced.

Cartridges and columns for purification of Strep-tag proteins with HPLC/FPLC, syringes or with peristaltic pumps


H-PR cartridges (Äkta compatible)	Pre-packed column with Strep-Tactin POROS^®.

H-PR cartridges: The H-PR cartridges are primarily designed for use with chromatography workstations with 10-32 connections (please note that you do not need adapters for HPLC and Äkta). They can, however, also be operated with other workstations, with syringes or with peristaltic pumps by use of common adapters for H-PR cartridges. Since column bodies are highly pressure resistant ("H-PR") and can tolerate 20 bar, the H-PR cartridges can be used with a flow restrictor. Cartridges can be connected in series to enlarge capacity. H-PR cartridges are available with Strep-Tactin Superflow^® (1 and 5 ml size) and Strep-Tactin Macroprep^® (1 and 5 ml size). They are also available with Ni-NTA resins.

Strep-Tactin POROS^® columns, pre-packed column with Strep-Tactin POROS^® 20 or 50, 1.7 ml bed volume, for HPLC/FPLC. Stable to high pressure (~170bar).

Adapters for cartridges

Adapters for H-PR cartridges are available for use with syringes and for connection to peristaltic pump chromatography systems or FPLC/HPLC workstations. Please note that no adapters are needed for connection of H-PR cartridges to Äkta systems or 10-32 connections.
